5,5-Dimethylhydantoin (1999)Abstract5,5-Dimethylhydantoin was, at worst, a mild skin and eye irritant in laboratory animals. It was a mild skin sensitizer in guinea-pigs. Low acute oral and dermal toxicity was demonstrated in laboratory animals. Studies involving repeated oral administration caused effects on the kidneys, and possibly the liver of rats, and tissue changes in the adrenals of mice. Oral administration to pregnant rats produced foetal toxicity at maternally toxic doses, and reproductive effects occurred when pregnant rabbits were exposed by the oral route. Rats treated orally showed no clear evidence of chromosomal damage or mutations. There was no evidence of mutagenicity in mammalian cells in culture, bacteria or yeast.
