1,1,1-Trimethylolpropane (TMP) is, at worst, a mild eye and skin irritant in laboratory animals. Patch tests evidently provoked no skin reactions in humans. TMP demonstrated low acute oral and dermal toxicity in laboratory animals. In limited studies involving repeated oral and inhalation exposure, effects were seen in the liver, spleen, heart, kidneys and lungs of rats. A summary noted no effects on reproduction in rats given repeated oral doses prior to mating, and during pregnancy and lactation. Brief reports claim no chromosome effects in mammalian cells in culture, and no evidence of mutagenicity in bacterial tests (including the Ames assay).
