Ethyl maltol was not an irritant to the skin of a rabbit. No skin irritation or sensitization was noted in volunteers treated with a dilute solution. Acute oral toxicity in rodents was moderate and acute dermal toxicity in a rabbit was low. In studies involving repeated oral administration to rats and dogs, effects were seen on the liver, kidney and blood. There was no evidence of carcinogenicity or reproductive toxicity in limited feeding studies in rats and dogs. Ethyl maltol was mutagenic in bacterial Ames tests but did not cause mutations in fruit flies or chromosome damage in the bone marrow of mice treated by injection.
