Hydroxypropylmethylcellulose has caused eye and skin irritation in humans and rabbits. Attempts to induce sensitization in guinea-pigs were unsuccessful. It was of low acute oral toxicity in rats. Repeated oral doses in rats and dogs caused diarrhoea and effects on the blood, kidney and liver. No evidence of carcinogenicity was seen in limited feeding studies in rats.
