Nigrosine was of low acute oral toxicity in the rat, although liver and kidney weights were altered and microscopic changes to the kidney and testes were reported following repeated oral administration. It failed to induce chromosomal damage in hamster cells in culture. No increase in tumour incidence was reported in a limited skin painting study in mice. A dilute solution was judged to be non-irritant to the rabbit eye and did not induce skin sensitization in guinea-pigs.
