Propyl gallate is a skin sensitizer and irritant in some individuals. The acute oral toxicity in laboratory animals was low. On repeated oral exposure tissue changes were seen in the liver, kidneys, stomach, forestomach and male reproductive organs in rats but not in mice. There was no conclusive evidence of marked reproductive toxicity in rats, rabbits, mice, hamsters or guinea-pigs following oral administration. Comprehensive long-term studies in mice and rats found no conclusive indication of carcinogenicity. Propyl gallate has not demonstrated any genotoxic activity in rats treated orally. Chromosomal damage and point mutations were induced in mammalian cells in culture but not in bacterial Ames tests.
